Turn the tap OFF and connect the other end which is a push fit to the low side port. That one you have shown is difficult to connect well to the low side without a leak though.
Low side is in the left of the engine bay.

Yes. It's got a push fit type of connector to connect to the car.
Pushfit connector: It takes a good push for it to click click on. If it doesn't make a great connection you loose refrigerant from the car which is not a good thing.
It isn't a professional bit of kit, but does the job if you are careful. Here it is on Amazon.co.uk

You can't beat a professional air con service by F1 autocentres. They have a machine that connects to both low and high side to diagnose faults before removing all the old air, moisture, oil and refrigerant and refilling the correct quantity of oil and refrigerant.
